Burke was the surname of a pure-blood[1] wizarding family, and one of the Sacred Twenty-Eight.
They were related to the Black, Flint[2] and possibly the Malfoy[3] families.
Caractacus Burke co-founded the Knockturn Alley shop Borgin and Burkes, which specialised in Dark artefacts, and it is possible that younger members of the family also became co-owners of the shop.
